of enzymes, including those
required for the oxidation of
pyruvate and fatty acids and for the
citric acid cycle.
Inner membrane. Folded into
numerous cristae, the inner
membrane contains proteins that
carry out the oxidation reactions of
the electron-transport chain and
the ATP synthase that makes ATP in
the matrix.
Outer membrane. Because it
contains a large channel-forming
protein (called porin), the outer
membrane is permeable to all
molecules of 5000 daltons or less.
Intermembrane space. This space
contains several enzymes that use
the ATP passing out of the matrix to
phosphorylate other nucleotides.

VÍA INTRÍNSECA • SE INICIA ANTE DAÑOS IRREPARABLES EN EL ADN CELULAR O POR AUSENCIA DE FACTORES DE SUPERVIVENCIA PRODUCIENDO LA SALIDA MITOCONDRIAL DE: – CITOCROMO C: SE UNE A APAF-1 (apoptotic protease activating factor-1) Y FORMA EL APOPTOSOMA QUE ACTIVA A LA CASPASA 9. – Smac/DIABLO: PROTEÍNAS QUE SE UNEN A IAPs (proteínas inactivadoras de la apoptosis) IMPIDIENDO SU EFECTO ANTI-APOPTÓTICO. VÍA INTRÍNSECA - La regulación de la apoptosis en esta vía es debida a la familia de Bcl-2 (B cell leukemia 2). - Bcl-2 fue descubierto como un proto-oncogén en linfocitos B malignos (linfoma de células B). - Existen 19 miembros que se dividen en tres grupos. - Cada miembro posee al menos uno de los cuatro motivos conservados denominados dominios de homología con Bcl-2 (BH): BH1-BH4. FAMILIA DE Bcl-2 1. Grupo I: - Actividad anti-apoptótica (Bcl-2, Bcl-XL , Bcl-w). - 4 motivos BH: BH1-BH4 - Prot. integrales de mb (cola hidrofóbica en el C-t) evitando la permeabilidad de la mb (mitocondria, RE). 2. Grupo II: - Actividad pro-apoptótica (Bax, Bak, Bok) - Carecen de BH4 3. Grupo III: - Actividad pro-apoptótica (Bid, Bik, Bad, Bim) - Sólo poseen el dominio BH3 - Pueden o no tener región transmembrana -Aumentan la permeabilidad formando poros en la mb (salida de citocromo C). CONTROL DE LA APOPTOSIS POR LA FAMILIA DE Bcl-2 - Los miembros anti-apoptóticos de la familia de Bcl-2 intentan bloquear el efecto pro-apoptótico de los otros. - Se forman heterodímeros de miembros pro y anti-apoptóticos neutralizándose hasta que algún estímulo altera ese equilibrio.
